MBPtag (maltose binding protein tag) can reduce the degradation of target proteins, increase the expression and stability of protein, and improve the water solubility of expression products, but the label is larger, which will have certain influence on the structure and function of protein.
Maltose binding protein (MBP) is a useful affinity tag that increases the level of expression and solubility of the protein of interest. The MBP tag also promotes proper folding of the attached protein. Because MBP can increase solubility, this tag is particularly useful for recombinant proteins that are aggregated in insoluble form (inclusion bodies). Affinity purification occurs under physiological conditions and is gently eluted using maltose. Mild elution retains the activity of the MBP-tagged protein.
Purification of the MBP fusion protein can be carried out under physiological conditions, using mild elution with maltose. Whether the MBP Tag is at the N- or C-terminus of the protein, the purification is very good. Dextrin is a ligand used in affinity purification of MBP fusion proteins. Uniform fine dextrin-conjugated Beads ensure that the MBP fusion protein elutes in a narrow peak, thus avoiding additional purification requirements. PurKineTM MBP-Tag Purified Fillers are also available in pre-packed columns and kits for added convenience. PurKine maltose binding protein tag dextrin resin (6% cross-linking) can be reused without regeneration, but with the increase of non-specifically bound proteins and protein aggregation, the flow rate and binding capacity are often reduced. The resin is cleaned to restore performance. Samples are recommended to be centrifuged or filtered through a 0.22μm or 0.45μm filter prior to loading to reduce impurities, improve protein purification efficiency and prevent plugging of the column. The product is based on a highly crosslinked 6% agarose gel and withstands pressures up to 0.3 MPa. It can be combined with peristaltic pumps and AKTA instruments for industrial purification.
